I dont believe thier is a specified break in period like there is for an engine, but 500 miles should be plenty of time to allow everything to seat properly.

according to s noted syty builder there is no break in, take it out make sure it shifts fine and then check fluid. When mine was rebuilt we ran it on a stand filled it up, took it out seemed like it was never going to shift to 2nd finally did, we pulled over to check fliud added a quart drove to around 100mph, went through the gears then we stopped did a 14psi launch and called it a day.

Let the transmission get up to operating temp *just* driving around (generally 15 minutes worth of driving will surfice). Then park it and do it two more times after it completely cools. Heat cycled mine over two days for my most recent rebuild. "Just driving around" means partial throttle at times but no WOT blasts and definitely NO boosted launches. The second part is hard to resist with a new trans but, it is for the best.
Change the fluid at around 1000 miles. Use Dex III with a bottle of Lubeguard. Get an add'l cooler if you don't have one. Derale pan or stacked plate cooler, or both.
I'd boost launch it once, get the seals to seat. Then take it easy for a couple days.
Most important thing is to make sure the TV cable is set correctly.
Drive along at about 20 percent throttle and take note of the MPH the shift changes occur. If you 1-2 shift isn't happening around 11-15MPH then you need to address that.

When I built my trans 1 1/2 years ago I didnt break it in. Did a boost launch 10 minutes into my driving and havent had a problem to this day(knock on wood). From being inside the trans myself, I dont really see what there is to break in. Course I dont so it for a living either but the test of time never lies either.
